盐酸吗啡缓释片【通用名称】盐酸吗啡缓释片【其他名称】盐酸吗啡缓释片 盐酸吗啡缓释片 拼音名：Yansuan Mafei Huanshipian 英文名：MOrphine Hydrochloride Sustained-release Tablets 书页号：2000年版二部-588 本品含盐酸吗啡(C17H19NO3.HCl.3H2O)应为标示量的93.0％～107.0％。 【性状】本品为薄膜衣片，除去薄膜衣后显白色。 【鉴别】取本品，除去薄膜衣，研细，取细粉适量(约相当于盐酸吗啡20mg)，置乳钵中，加水 10ml，研磨 使盐酸吗啡溶解，滤过，滤液蒸干，残渣照盐酸吗啡项下的鉴别(1)、(2)、(3)、(5)项试验，显相同 的反应。 【检查】含量均匀度 取本品1片，置乳钵中，加水适量，研磨使盐酸吗啡溶解，转移至100ml (10mg规格) 或250ml(30mg规格)、500ml(60mg规格)量瓶中，用水分次洗涤乳钵，洗液并入量瓶中，充分振摇，加 水稀释至 刻度，摇匀，用0.45μm滤膜滤过，取续滤液，照含量测定项下的方法，自“精密量取续滤液”起，依 法测定含量，应符合规定(附录Ⅹ E)。 释放度 取本品，照释放度测定法(附录Ⅹ D 第一法)，采用溶出度测定法第一法装置，以水 500ml为溶 剂，转速为每分钟50转，依法操作，经1小时、2小时、3小时、4小时、5小时和6小时时，分别取溶液 2ml，用 0.45μm滤膜滤过，并及时在操作容器中补充相同温度的水2ml，取续滤液作为供试品溶液。另精密量 取含量 测定项下的对照品溶液适量，分别加流动相稀释成每1ml中含0.02mg(10mg规格)、0.05mg(30mg规 格)、 0.1mg(60mg规格)的溶液作为对照品溶液，照含量测定项下的方法分别测定峰面积，计算出每片在不同 时 间的释放量。本品每片在1小时、2小时、3小时、4小时、5小时和6小时时的释放量应分别相应为标示 量的 25％～45％、40％～60％、55％～75％、65％～85％、70％～90％和80％以上，均应符合规定。 其他 应符合片剂项下有关的各项规定(附录Ⅰ A)。 【含量测定】照高效液相色谱法(附录Ⅴ D)测定。 色谱条件与系统适用性试验 用十八烷基硅烷键合硅胶为填充剂；以0.05mol/L磷酸二氢钾溶液- 甲 醇(4:1)为流动相；检测波长为280nm。理论板数按吗啡峰计算应不低于1000。 测定法 取本品10片，精密称定，研细，精密称取适量(约相当于盐酸吗啡35mg)，置250ml量瓶 中，加 水适量，充分振摇使盐酸吗啡溶解，加水至刻度，摇匀，用0.45μm滤膜滤过，精密量取续滤液20μl注 入液相 色谱仪，记录色谱图；另精密称取经80℃干燥至恒重的吗啡对照品适量，加流动相制成每1ml中含 0.1mg的 溶液，同法测定。按外标法以峰面积计算，结果乘以1.317，即得供试品中C17H19NO3·HCl·3H2O的 量。 【类别】
