硫酸吗啡控释片【通用名称】硫酸吗啡控释片【其他名称】硫酸吗啡控释片 硫酸吗啡控释片 拼音名：Liusuan Mafei Kongshi Pian 英文名：Morphine Sulfate Controlled-release Tablets 书页号：2000年版二部-865 本品含硫酸吗啡[(C17H19NO3)2.H2SO4.5H2o]应为标示量的95.0％～105.0％。 【性状】本品为薄膜衣片，除去薄膜衣后显白色。 【鉴别】取本品的细粉适量(约相当于硫酸吗啡0.1g)，加水10ml振摇使硫酸吗啡溶解，滤过，滤 液做以下试验。 (1)取滤液0.5ml，加甲醛浓硫酸溶液(取甲醛0.5ml，小心加入浓硫酸9.5ml)1ml，即显紫堇色。 (2)取滤液0.5ml，加水5ml，加铁氰化钾试液1滴、三氯化铁溶液(取三氯化铁5g，加水100ml溶 解)1滴，即显蓝绿色，立即变为蓝色。 (3)取滤液5ml，加2mol/L盐酸溶液1ml、氯化钡溶液(取氯化钡细粉6.1mg，加水100ml溶解) 1ml，即产生白色沉淀，加碘-碘化钾溶液(取碘2.0g、碘化钾3.0g，加水100ml溶解)1滴，即产生黄色 沉淀 ,再加氯化亚锡溶液(取氯化亚锡33.0g，加2mol/L盐酸溶液10ml溶解，加水至100ml)2～3滴，煮 沸，沉淀颜色消失。 【检查】有关物质 取含量测定项下的本品细粉，精密称取适量(约相当于硫酸吗啡20mg)，加氢 醌溶液(取氢醌30mg，加流动相1000ml溶解)5ml，在水浴中超声处理，使硫酸吗啡溶解后，摇匀，滤 过，取续滤液作为供试品溶液；另精密称取硫酸吗啡对照品适量，加氢醌溶液溶解并定量稀释制成每 1ml中约含80μg的溶液，作为对照品溶液。照高效液相色谱法(附录Ⅴ D)测定，用十八烷基硅烷键合硅 胶(5μm)为填充剂；甲醇-庚烷磺酸钠醋酸溶液(取庚烷磺酸钠2.02g，加水适量溶解，加冰醋酸5ml， 加水稀释至1000ml，摇匀)(70:90)为流动相；检测波长为233nm。理论板数按硫酸吗啡峰计算应不低 于1000。量取对照品溶液5μl注入液相色谱仪，调节仪器检测灵敏度，使主成分吗啡峰高度约为记录仪 满量程的20％～30％，再淮确量取对照品溶液和供试品溶液各5μl，分别注入液相色谱仪，记录色谱图 至主成分吗啡峰保留时间的4倍。供试品溶液色谱图中如有杂质峰，各杂质峰面积总和不得大于对照品 溶液主成分吗啡峰面积。 含量均匀度 取本品1片，研细，用氢醌溶液定量转移至50ml量瓶中，在水浴中超声处理，使硫酸 吗啡溶解后，加氢醌溶液稀释至刻度，摇匀，滤过，取续滤液2ml，置25ml量瓶中，加氢醌溶液稀释至 刻度，摇匀，作为供试品溶液；另精密称取硫酸吗啡对照品，加氢醌溶液溶解并定量稀释制成每1ml中 约含48μg的溶液，作为对照品溶液。取上述两种溶液，照含量测定项下的液相色谱条件测定，并计算 出每片的硫酸吗啡含量，应符合规定(附录Ⅹ E)。 释放度 取本品，照释放度测定法(附录Ⅹ D 第一法)，采用溶出度测定法(附录Ⅹ C 第二法) 的装置，以 pH6.5磷酸盐缓冲液900ml为溶剂，转速为每分钟100转，依法操作，在1小时、2小时、3小时、4小 时、5小时和6小时分别取溶液5ml，用0.8μm微孔滤膜滤过，并即时在操作容器中补充pH6.5磷酸盐缓 冲液5ml，取续滤液作为供试品溶液；另精密称取硫酸吗啡对照品适量，用上述缓冲液制成每1ml中约 含30μg的溶液，作为对照品溶液。分别取上述两种溶液各10μl，照含量测定项下的液相色谱条件测 定，依次注入液相色谱仪，记录色谱图，按外标法以峰面积或峰高分别计算出每片在不同时间的释放 量。本品每片在1小时、2小时、3小时、4小时、5小时和6小时的释放量应分别相应为标示量的30％～ 45％、45％～65％、55％～75％、65％～85％、75％～95％、和80％以上，均应符合规定。 其他 应符合片剂项下有关的各项规定(附录Ⅰ A)。 【含量测定】照高效液相色谱法(附录Ⅴ D)测定。 色谱条件与系统适用性试验 用十八烷基硅烷键合硅胶(5μm)为填充剂；以甲醇-庚烷磺酸钠醋酸 溶液(取庚烷磺酸钠2.02g，加水适量溶解，加冰醋酸5ml，加水稀释至1000ml，摇匀)(50:50)为流动 相；检测波长为233nm。理论板数按硫酸吗啡峰计算应不低于1000。 测定法 精密称取硫酸吗啡对照品约30mg，置50ml量瓶中，加氢醌溶液溶解、并稀释至刻度，摇 匀，精密量取2ml，置25ml量瓶中，加氢醌溶液稀释至刻度，摇匀，作为对照品溶液；另取本品20片， 精密称定，研细，精密称取适量(约相当于硫酸吗啡30mg)，置50ml量瓶中，加氢醌溶液，在水浴中超 声处理，使硫酸吗啡溶解，加氢醌溶液稀释至刻度，摇匀．滤过，精密量取续滤液2ml，置25ml量瓶 中，加氢醌溶液稀释至刻度，摇均，作为供试品溶液。精密量取对照品溶液与供试品溶液各10μl，分别 注入液相色谱仪，记录色谱图，按外标法以峰面积计算，即得。 【类别】麻醉镇痛药。 【规格】30mg 【贮藏】遮光，密封保存。
